Design Notes

The design of this part considered the structural integrity required for proper receptor function, focusing on the accurate representation of the receptor's seven-transmembrane domain. Special attention was given to the receptor's ligand binding site, particularly the ECL2 region, which plays a crucial role in ligand specificity and receptor activation. Challenges such as ensuring correct membrane localization and effective coupling with Gi/o proteins were addressed to optimize the receptor's performance in synthetic biology applications.


Source

The MT2 receptor sequence is derived from mammalian species, with its gene sequence originally cloned from human tissues. It is a well-characterized GPCR, playing a significant role in the regulation of melatonin's effects in the brain and other peripheral tissues.
